Comparison of Immune System of Newborns and Adults
Dusilová, Adéla ; Hrdý, Jiří (advisor) ; Zajícová, Alena (referee)
In general, it is possible to characterize neonatal immune system (IS) as immature in comparison to adult IS. From a clinical point of view, newborns show an increased susceptibility to infections. Breastfeeding can contribute to the descent incidence of illnesses, because it supplies the intestinal mucosal system with antibodies of the mother`s origin, important nutrients and other immunoregulatory components. Breast milk compensates decreased newborn's capacity to produce immunoglobulins- especially IgA, that concentration reaches adult levels in two years, but even later (to the pubescent period). Other classes of antibodies are found in cord blood only sporadically except IgG, which is transferred transplacentary. Reduced ability of B lymphocytes to produce antibodies is caused by insufficient expression of surface costimulatory signals of Th2 cells. T lymphocytes are not able to react properly to low doses of stimulators (polyclonal activators - phytoid lectins: ConA or PHA), which bind to T cell receptors in complex with CD3 and proliferate in a response to anti-CD3 monoclonal antibodies. Most of the cord blood T lymphocytes display "naive" phenotype CD45RA. During intrauterine development, neonatal IS is in contact with mother IS and because a pro-inflammatory Th1 response could lead to...
Preparation of affinity surfaces for clinical diagnostics
Kalaninová, Zuzana ; Pompach, Petr (advisor) ; Žáček, Petr (referee)
Botulinum toxins are group of protein venoms produced by Clostridium bacteria. These toxins belong to the most toxic substances causing paralytic disease called botulism. The endopeptidase activity of the toxins leads to the blockage of neurotransmitters release to neural junctions. Since the first symptoms of botulism are observed within 48 hours and toxin remains active for months, botulinum toxins are considered as potential bio- terrorism agents. For successful treatment, fast assays for detection of the toxin presence and type are highly required. This work presents a new method that could be potentially used for determination of botulotoxin in various matrices. The method is based on the detection of products of botulinum toxin A (BoNT/A) endopeptidase activity by affinity surfaces prepared by ambient ion soft landing and MALDI mass spectrometry. For immobilization and subsequent enrichment of biotinylated products of BoNT/A activity, proteins with high affinity to biotin - avidin, neutravidin and streptavidin - were used. The presented method, which combines affinity surfaces with MALDI mass spectrometry is sensitive, robust and fast and might be potentially used for detection of BoNT/A activity. Key words: ambient ion soft landing, neutravidin, botulinum toxin A, BoNT/A, MALDI/TOF mass...
Fear in The Space
Savincová, Karolína ; Pfeiffer, Jan (advisor) ; Gajdošíková, Pavla (referee)
The thesis deals with the concept of fear in space. Through a/r/tographic research, the author examines the concept and changes in its perception during the process of creating an artistic and pedagogical work. In the first part, the author documents theoretical and methodological framework of the research, designs the research proposal and defines terms related to it. In the second part, the author presents her own way of grasping the concept in the space and introduces the concepts that related with this topic. She researches the concept through the reflection of her own work, both artistic and then pedagogical. As part of her artistic work, the author acquaints the reader with the origin of the artwork and follow-up reflection, from which comes the creation of visual task. In the third part, she analyses the primary documents using three-phase coding and then creates concept maps from the obtained codes, which are used to visualize the display of research results. Interprets the data obtained from the research. In the fourth part, the author summarizes the results of research, answers research questions and evaluates the achievement of the research goal. From the results of the analysis of available documents and by comparing conceptual maps, she finds out whether the understanding of the...
The Phenomenon of Architecture in One's Own Art and Pedagogical Work (Pedagogical Implications)

Pavla Gajdošíková: The Phenomenon of Architecture in One's Own Art and Pedagogical Work (Pedagogical Implications) Abstract: The dissertation looks at the perception of architecture and space in art and explores the possible uses of artistic and reflective processes in learning situations involving students of art education. It is based on ongoing public discussions regarding architecture's influence on the perception of the environment and the associated quality of life of each and every one of us. Thematically, it follows on the author's cycle of artworks titled Subjective Urbanism, which explored the subject using various artistic media. The dissertation makes use of a/r/tography, a methodology that offers a way of understanding a subject of research from three different perspectives (artist, teacher, researchers), thus enabling complementarily structured learning in "interspace." Metaphorically speaking, the subject of architecture and space is approached as an intertwining of these three identities. This also the paper's triadic structure. In the first part, the author introduces the subject at hand through her own works of art, a reflection of which formed the starting point for her subsequent inquiries. In the second part, she uses several case studies to describe three pedagogical projects aimed at...

Effects of message design and content on the performance of email marketing campaigns
Miadoková, Miroslava ; Koudelková, Petra (advisor) ; Huněk, Filip (referee)
Diploma thesis deals with the influence of the selected attributes of e-mails. Based on the previous research and results they should change the behaviour of the recipients. In the theoretical part author will explain the e-mail and its issues as one of the tools used in new media. The closer focus will be given to the optimalization of e-mails as it will be used for the basis of research. The practical part identifies what features of content and design of email marketing significantly influence the level of public engagement. The methodological basis of research is quantitative method. To test the results of hypotheses a technique of A/B testing will be used. The test focused on selected features will be sent and monitored on the sample of Czech subscribers of the cosmetic company. The results will be measured using the click through rate and the value of the purchases made.
